Principal Scientist/Associate Director, Vector Development (AD06-2)



TScan Therapeutics is a startup biotech seeking to revolutionize life-changing therapeutics for patients by identifying novel T cells and epitopes. We are looking for a Principal Scientist / Associate Director, Vector Development & Ex Vivo T Cell Engineering to lead efforts in developing ex vivo genetic modification tools for the company’s novel TCR-engineered T cell therapy products.  This role will reside within CMC and will report to the Associate Vice President of Process & Analytical Development. Key activities will include leading efforts related to non-viral vector design & testing, screening and implementation of gene editing tools, bench scale cell process feasibility studies, tech transfer to process development, and acting as a SME for external vendors.

The ideal candidate will (i) have a strong background in molecular cloning, ex vivo gene editing & modification, and cell therapy product development; (ii) be a steadfast problem solver who will thrive in a fast-paced, lean, and agile environment; and (iii) be a team player who can foster productive relationships with internal colleagues and with external partners.

The ideal candidate will be a technical leader who is well-versed in molecular cloning, design and testing of functional vector elements, CRISPR-based gene editing techniques, and cell-based analytics.  Experience with DoE-based experimental design and developing cell therapies for immuno-oncology indications is highly desired. Prior experience in protein expression / purification strategies is a plus.


TScan Therapeutics was founded in 2018 by a small group of scientists led by Dr. Stephen Elledge (Harvard Medical School/HHMI). TScan is discovering and developing novel TCR-engineered T cell therapies for the treatment of cancer. TScan’s discovery platform is centered around a breakthrough technology that enables the rapid, comprehensive, and genome-wide identification of the peptide antigen targets of T cell receptors. This technology greatly enhances our ability to understand the specific antigens that drive important disease processes, including tumor cell recognition by the immune system, self-reactivity in autoimmune disorders, and acquired immunity in infectious disease. TScan is using the platform in oncology to discover new TCR/target pairs and to rapidly move novel TCRs into clinical development.


  • As a senior leader within the Process & Analytical Development Team, provide strategic, technical, and scientific leadership on vector development and cell engineering strategy
  • Develop and drive experimental project plans, providing training and oversight to junior staff
  • Work cross-functionally with discovery and pre-clinical teams in developing vector elements for lead candidates
  • Foster a culture of continuous improvement and scientific collaboration
  • Oversee process and method transfer and provide technical support to PD and AD as needed
  • Provide technical guidance to CMC team and act as the technical liaison for external vendors
  • Collaborate closely with other key line functions and communicate complex data/decisions within the department and to program teams


  • Principal Scientist: PhD with 8+ years of relevant experience; Associate Director: PhD with 10+ years of relevant experience. Degree emphasis in biological sciences, biochemistry, cell biology or related field is preferred
  • Strong background in molecular biology, vector development, and experience with CRISPR-based gene editing
  • Previous experience with transposon-based gene integration is highly desired
  • Scientific creativity, critical thinking, ability to innovate, and analytical problem solving are a must
  • Excellent communication skills (both written and oral)
  • Experience drafting manufacturing documentation and supporting documentation for regulatory filings
